In making wines, grape juices serve as the source of both macronutrients and micronutrients required by the microbes, while beer making utilizes seeds. A cost/benefit analysis led to this difference. After it became evident that the seeds used in making beer were critical sources of food for animals, it proved rational for grapes serve as food for microbes in the wine making process, preserving the grains for animals (14). Though this may seem harmless on the outside, these opinions have become the poison apple of today’s culture, tearing teenagers lives apart. Specifically, teenagers are falling to eating disorders due to the pictures and habits in the everyday community. Large billboards of flawless models, magazines with dieting tips and get-skinny schemes flood the minds of teenage girls and boys, imprinting the thought that they are obligated to look perfect. Eating disorders have very serious health risks, which can be aided by extensive treatment. Specific habits and signs can identify each disorder. The signs of anorexia nervosa and bulimia are vastly different. Girls that suffer from Anorexia tend to be perfectionists who want to control the amount of food consumed, where Bulimics tend to eat large amounts of food and purge the calories consumed. In anorexia, girls will start â€Å"Wearing baggy clothing, or clothing inappropriate for the season (long sleeves on a hot day)† (Is Your Child Showing Signs of an Eating Disorder?, PR Newswire). This is due to their need to hide the extreme amount of weight loss. Bulimics will hide their purges by spending a lot of time in the bathroom and running the water to cover the sound. Girls with these disorders are sneaky and manipulative. They will go great lengths to hide what they are doing. Although the person that is affected will be in denial these signs are easy to spot and more noticeable then they think. Some times these signs are what can save a persons life. Anorexia Nervosa and Bulimia are detrimental to the human body. Anorexia can lead to dizzy spells, irregular menstrual cycles, hair loss and breakage and eventually death. Bulimia can lead to damaged teeth, rips in esophagus, sores in the mouth and eventually death as well. The health risks are beyond physical but mental as well. Girls with eating disorders almost always have another psychiatric disorder. Some can suffer from depression and anxiety. Eating disorders are often classified as a â€Å"Dual Diagnosis†. With the correct treatment, both disorders can be reversed. It is important for the patient to recognize their symptoms and realize that they are not alone in their situation. Parental support is normally the most important factor in recovery â€Å"The most important thing a parent can do is listen, says Susan McClanahan, Ph.D., a clinical psychologist who specializes in the treatment of anorexia and bulimia.† Treatment is the best answer and is an important step to recovery. Recovery is a slow process that needs a lot of understanding and patience. If the treatment is rushed the patient is more likely to fall back into old habits and relapse. Some cities in south-eastern Australia had over 15500 km-2 of pl astic concentrations. During the a long term of flowing, these plastics could be broken down into small fragments within 5 mm length, which commonly called microplastic and damaged to the food web and ecosystem easily. Therefore, we had two methods to solve this issue. Approximately 37% of plastics were used for manufacturing disposable packaging, comprising bottle, cups and bags,hence, the first solution was reducing the production of single-use plastic package and decreased the demand of using these plastic manutacture by advocating awareness of this water issue (Schneider J 2013). Via this solution, Australia could have sufficient supply and overcame water scarcity lightly. But on the contrary, cloud seeding can be very costly. In most of the cases, it takes millions of RMB to implement one cloud seeding, therefore, Australia would better have a strong economic background beforehand like developing their agriculture or IT industry. On the other hand, as for the water pollution in Australia, most of water is polluted by fragmented plastic and majority of people and factories concentrated in the eastern coast of the Australia, so we could see eastern Australia is more polluted than any other part of Australia. Moreover, because it is hardly to eliminate all these plastic pollutants and made sure there is no plastic contamination in the furture in a scientific way, we could just do it in a political approach. For instanc e, government could enact laws of environmental protection or making some advertisments to persuade people do not throw plasctic bags or products into any kind of natural water source. In addition, some eco-friendly organizations can just simply set some dustbins around those highly polluted cities for recycling the plastic. But its a little bit tough for them to conquer water pollution via this method. All in all, I think Australia can conquer these two water issues based on the previous contents. 5.0 Conclusions and Recommendations In general, cloud seeding still got a substaintia chance to resolve water restriction by it extra precipitation. Australia just need to develop their economy and have enough capital to conduct these experiments. To handle the increasing plastic pollution in Australian water, government have appeal people do not throw any plastic manufactures casually and set several laws to forbid people from doing this, in particular the eastern part of Australia. The effectiveness of this measure rely on the governmental actions and Australian qualities. There are some limitations in this scientific paper, first of all is source restrictions. Why do you believe that John Brown believed that the situation in the U.S. at the time could only be solved by bloodshed and not compromise?   Ã‚  Ã‚  Ã‚  Ã‚  I believe that there are many reasons why John Brown believed that violence was the only way he could prevail in the fight to end slavery in the United States. First of all; at this time in history, issues were moving fairly quickly. As soon as Kansas was to become a territory, supporters from both the North and South rushed to settle on the land and tried to gain control of the vote in the area. Secondly, it probably seemed that any attempts at compromise at that time had not, and would not, get very far in the issue. Lastly, the most prominent reason I believe that John Brown felt violence was the only answer, was because of his belief in God. As he stated in his speech to the courts during his trial, he felt that what he did was not wrong. Sacrificing his life was worth it; even if it was just one step towards ending slavery, something that was so cruel and wicked anyway, his violence was no worse than the slavery.   Ã‚  Ã‚  Ã‚  Ã‚  When the Kansas-Nebraska Act was introduced in the U.S., the North was upset because the new territories would probably be pro-slavery. As soon as they could, both sides of the issue sent in settlers to try and gain control. This â€Å"race for Kansas† made the race to make a final decision on the issue of slavery in the U.S. an even more urgent issue. This battle to decide what side the two new territories would be on probably pushed the issue so hard it caused a lot of discomfort and probably hurried people like John Brown into making decisions quickly, because people were afraid. I believe that the rush to make a ruling for or against slavery was one reason why John Brown chose bloodshed over compromise.   Ã‚  Ã‚  Ã‚  Ã‚  The second reason I believe Brown picked violence over negotiation was because of the lack of communication. It probably seemed a hopeless endeavor to even try to correspond with the South on the issue, especially after violence had already begun in many places; not to mention in the act of slavery itself. It would have been very hard for either side to civilly speak or negotiate with the opposition in an reasonable manner.

I. Theory of Existentialism Existentialism is a philosophical movement that posits that individuals create the meaning and essence of their lives, as opposed to deities or authoritites creating it for them. It emerged as a movement in twentieth-century literature and philosophy, though it had forerunners in earlier centuries. Existentialism generally postulates that the absence of a transcendent force (such as God) means that the individual is entirely free, and therefore, ultimately responsible. It is up to humans to create an ethos of personal responsibility outside any branded belief system. In existentialism views, personal articulation of being is the olny way to rise above humanity`s absurd condition of much suffering and inevitable death. Existentialism is a reaction against traditional philosophies, such as rationalism and empiricism, that seek to discover an ultimate order in metaphysical principles or in the structure of the observed world, and thereby seek to discover universal meaning. Existentialism originated with the nineteenth-century philosophers Soren Kierkegaard and Friedrich Nietzsche. It became prevalent in Continental philosophy, and literary figures such as Fyodor Dostoevsky also contributed to the movement. In the 1940s and 1950s, French existentialism such as Jean-Paul Satre, Albert Camus, and Simone de Beauvoir, wrote scholarly and fictional works that popularized existential themes such as â€Å"dread, boredom, alienation, the absurd, freedom, commitment, and nothingness. † Walter Kaufmann describes existentialism as â€Å"The refusal to belong to any school of thought, the repudiation of the adequacy of any body of beliefs whatever, and especially of systems, and a marked dissatisfaction with traditional phylosophy as superficial, academic, and remote from life. † Existentialism tends to focus on the question of human existence – the feeling that there is no purpose, indeed nothing, at the core of existence. Finding a way to counter this nothingness, by embracing existence, is the fundamental theme of existentialism, and the root of the phylosophy’s name. In existentialism view, they asserts that a human finds oneself already in a world and prior context that the human cannot think away. In other words, the ultimate and unquestionable reality is not consciousness but existence. A central proposition of existentialism is that humans define their own meaning in life. Such a view might be phrased technically by philosophers as existence precedes essence, that is a human’s existence conceptually precedes the essence or meaning that may be ascribed to the life. Satre, in Essays in Extentialism, further highlights this consciousness of being thrown into existence in the following fashion: â€Å"If man, as the existentialist conceives him, is indefinable, it is because at first he is nothing. Only afterward will he be something, and he himself will have made what he will be. † Emphasizing action, freedom, and decision as fundamental, existentialist oppose themselves to rationalism and positivism. That is, they argue against definitions of human beings as primarily rational. Rather, existentialists look at where people find meaning. Existentialism asserts that people actually make decisions based on what has meaning to them rather that what is rational. A further type of existentialism is agnostic existentialists, who make no claim to know whether or not there is a â€Å"greater picture†; rather, they simply assert that the greatest truth is that which the individual chooses to act upon. II. What is Superhero A superhero – sometimes written as super hero – is a fictional character of extraordinary physical ability dedicated to acts in the sake of public interest. Since the debut of the prototypal superhero Superman in 1938, stories of superheroes – ranging from brief episodic adventures to continuing years-long sagas – have dominated American comic books and crossed over into other media. A female superhero is sometimes called a superheroine or super heroine. By most definitions, characters need not have actual superhuman powers to be deemed superheroes, not, although sometimes terms such as costumed crimefighters are used to refer to those without such powers who have many other common traits of superheroes. The two-word version of the term is a trademark co-owned by DC Comics and Marvel Comics. There have been successful superheroes in other countries most of whom share the conventions of the American model. Examples include Cybersix from Argentina, Captain Canuck from Canada and the heroes of AK Comics from Egypt. For this Western area, I will take Batman as the representative. Unlike many superheroes, Batman has no superpowers and instead relies on his own scientific knowledge, detective skills, an athletic abilities. Batman is physically at the peak of human ability in dozens of areas, notably martial arts, acrobatics, strenght, and escape artistry. Rather than simply outfighting his opponents, Batman often uses cunning and planning to outwit them. Batman’s costumes incorporates the imagery of a bat in order to frighten criminals. Japan is the only country that nears the US in output of superheroes. The earlier of these wore scarves either in addition to or as a substitute for capes and many wear helmets instead of masks. Moonlight Mask, Ultraman, Kamen Rider, Super Sentai (the basis for Power Rangers), Metal Heroes and Kikaider have become popular in Japanese tokusatsu live-action shows, and Science Ninja Team Gatchaman, Casshan, The Guyver, and Sailor Moon are staples of Japanese anime and manga. However, most Japanese superheroes are shorter-lived. While American entertainment companies update and reinvent superheroes, hoping to keep them popular for decades, Japanese companies retire and introduce superheroes more quickly, usually on an annual basis, in order to shorten merchandise lines. In addition, Japanese manga often targets female readers, unlike U. S. comics, and has created such varieties as â€Å"magical girl† (e. g. Cardcaptor Sakura) for this audience. For this Eastern area, I will take Naruto as the representative. Naruto whose full name is Uzumaki Naruto is a young boy who dreams of becoming the leader of his Hidden Village. It will be difficult though for Naruto, because when he was a baby an evil demon was placed inside him to stop its rampage. As Naruto grew the townspeople saw Naruto himself as the demon, even though he was merely its container. Naruto lives in a world populated by ninja villages. The ninja serve as the armies for the countries that inhabit the world. Most of the countries have their own Hidden Village, which serves to train and manage the ninja of the country. Also the ninja in the series are able to utilize jutsu techniques, which are the secrecy, body and illusion arts of the ninja. These allow the ninja to perform many amazing skills such as the manipulation of the surrounding elements. III. Batman and Naruto: Their Differences and Their Revelance In Existentialism The true feature of Batman as a common people is Bruce Wayne, a millionaire industrialist. He inherited a big corporation, Wayne Corporation, from his father. The ego ‘Batman’ was born because of two accident. The first was the murder of Bruce’s parents. Bruce witnessed his parent killed in a gun-accident. A street-criminal shot them to death. The second was Bruce falling down into a cave in Wayne’s manor. He fell into a dark and humid cave where a hundred of bats lived. Those two accidents led Bruce to learn about martial arts and gadget-operating skill in order to fight the crime in Gotham city. His fear of bat made him wear a bat-costume in doing his operation. Thus, Batman as a superhero who fight a crime has born. This is the very purpose why Batman wants to fight crime: a personal vendetta against criminals. Since Batman does not have any superpowers, he relies on his own scientific knowledge, detective skills, an athletic abilities. Also his wealth enables him to get much modern and sophisticated gadget in order to beat his nemesis. In other hand, Naruto, instead of his lack of intellectual ability in solving a problem, his friend still considers some of his actions brilliant. Naruto does all of his action by intuition, not by a precise step as a ninja should do. What makes Naruto a superhero is that he possesses massive abundant chakra energy inside his body. This chakra is originated from a Nine-Tailed Demon Fox that is sealed inside his body. Together with his friends and mentor, Naruto solve the problem and fight with the evil ninjas. They act in a group of 4 ,3 ninjas and a mentor. They perform a close combat by using martial arts and ninjutsu, and a long-range combat by using flying dagger or shuriken. Once they accomplished a mission, they report to the hokage, which is the leader of the village. Yet, the very important thing that makes they both become a superhero is that there exist some nemesis, villains, that must be eliminated. They both make some struggles to get rid of those system distracted. What differentiate them is the way they make these struggles. Batman eliminates crime in Gotham by his own rule. He does not obey the rules prevailed in that city. He intentionally takes action againts criminal without involving the authorities. It can be concluded that Batman makes his own system to fix the system. However, Batman – intentionally or not – does not kill his enemy. He just sents those criminals to the authorities, to be prisoned or not. Batman’s worst nemesis, The Joker, is still alive until now. Although they often have some ‘fight’, but neither Batman or The Joker is condemned to death. While Naruto’s job is to keep the system in his village to be kept save. He does not violate the system prevailed in his village, instead he is forced to obey that system. His acts are being ruled by Hokage, the highest authority in the village. He is a kind of ‘paid’ superhero that acts upon a mission that is given by that Hokage. Once they accomplished that mission, they have report to that Hokage. Here, Naruto biggest enemy, Kabuto, is remain alive until now. What makes this difference? It is because the different culture that affect the people in West and East. Why Batman have a nerve to make his own system to fix the main system prevailed in his city is due to the Western way of thinking. Western culture tends to emphasize critical thinking. They are learned to break the rules if they think that there are some errors in those rules. Their new ideas are being welcomed, even they are supported to make new inventions. While Eastern country tend to ‘close their eyes’. They have to accept what the older people says. They are not being couraged to make some critics toward the culture. They tend to give high respect (sometimes not in a proper measure) toward conventional ideas. Here it implies in the way Batman and Naruto makes their struggle to fight crimes. Then another point where existentialism takes place also emmerges. Why those superheroes do not kill their enemies? It is because they ‘need’ their nemesis so that they can be a superheroes still. Let’s we go back to the question of what makes those superheroes superhero? What do they pursue? They want to eliminate crimes. Then we come to the question of what causes this crime? Criminals. So superheroes exist because there are some criminals. If there are no criminals, so there are no superheroes. Like what Satre has said, â€Å"If man, as the existentialist conceives him, is indefineable, it is because at first he is nothing. Only afterward will he be something, and he himself will have made what he will be. † Thus, superhero is nothing if there is no criminal to make him defineable. This is the meaning of existence preceding essence. I would like to use the term that coined by Heidegger, â€Å"throwness†, that is human beings are â€Å"thrown† into existence without having chosen it. Whether superhero or the enemy do not have the power to choose what they want to be. First they just exist in the world, then they just do the thing that they believe as a greatest truth. More that 2,000 years ago the birth of one baby born in the tiny, dusty village of Bethlehem changed the world and divided history into BC and AD. It reminds us that we must feed on Christ. This child of Bethlehem (Bethlehem means house of bread) announced that He is the Bread of Life. By faith and repentance we spiritually eat of the Christ of Christmas. His torn flesh and spilt blood on the cross is our salvation and daily bread. Third BETHLEHEMS CELEBRATION. The night Christ was born an angel appeared to shepherds out in the field guarding their sheep. They announced the good news, Dont be afraid! I bring you good news of great joy for everyone! The Savior yes, the Messiah, the Lord has been born tonight in Bethlehem. Suddenly the angel was joined by a vast host of others the armies of heaven praising God. Glory to God in the highest heaven and peace on earth to all whom God favors' (Luke 2:8-20 NLT). For all those who receive Christ into their heart as Savior and Lord, there is a joyful celebration. Isaiah prophesied of this day, In that day you will sing . . . with joy will you drink deeply from the fountain of salvation! In that wonderful day you will sing (Isaiah 12 NLT). The average density of the aluminum cylinders was 4.29g/cm3, and the density of the zinc cylinders was 6.079g/cm3. Both of these values seem reasonable, and the zinc seems to be slightly heavier than the aluminum. The density of the zinc cylinders was found using the volume by displacement method, and the volume of the aluminum cylinders was found by the formula method V= (3.14d2h)/4. Problems with the equipment may have possibly introduced error into the measurements. The 100mL graduated cylinder used to find the volume of the zinc cylinders was only marked in 10mL increments, requiring estimation for levels in between. The volume of the aluminum cylinders required the use of the vernier caliper to measure the diameter and the length of each cylinder. Improper use of the vernier caliper may have altered the measurements received for the aluminum cylinders. The error introduced by these problems was slight, however, and the calculated densities for both objects are probably quite reliable.

Fe-AZT and Pd-AZT Synthesis and Effects Synthesis and Effects of Fe-AZT and Pd-AZT on Viability of Human Hepatocytes and Hepatocellular Carcinoma Cells Submitted by: Anna Harutyunyan Introduction Cancer is one of the major causes of mortality in the world. In 2015, according to the National Cancer Institute, over 1.6 million new cases of cancer were reported in the United States. The estimated cancer deaths for the year of 2015 were over 500,000. According to the National Cancer Institute projections, in 2016 an estimated 1.7 million new cases of cancer were diagnosed in the United States and 595,690 people will die from the disease (National Cancer Institute). Although new cancer treatments and therapies are designed and implemented every year, cancer is still the number two cause of mortality in the United States, therefore developing and testing new effective anticancer agents is crucial. Hepatocellular carcinoma (HCC) is one of the deadliest forms of cancers (Venook et al., 2010). The overall 5-year survival rate of HCC is less than 17%, making HCC the fastest rising cause of cancer related death in the United States (American Cancer Society 2016; Mittal and El-Serag, 2013). The annual age-adjusted incidence rates of HCC increased from 1.4 per 100,000 individuals in 1975-77 to 4.8 per 100,000 in 2005-07. An estimated 39,230 new cases of liver cancer (including intrahepatic bile duct cancers) were expected to occur in the US during 2016, approximately three-fourths of which would be hepatocellular carcinoma. An estimated 27,170 liver cancer deaths were expected in 2016 (American Cancer Society 2016). In 80-90% cases HHC occurs with and after cirrhosis. HCCs major risk factors are Hepatitis B and C viruses, cirrhosis, non-alcoholic fatty liver disease (NAFLD), overconsumption of alcohol and exposure to other carcinogenic substances (Mittal and El-Serag, 2013). Liver ca ncer is the sixth most common neoplasm and the third leading cause of cancer-related deaths, accounting for approximately 600,000 deaths annually (Venook et al., 2010). Because of early metastasis and progression, HCCs treatment is difficult, and traditional chemotherapy has shown limited success (Sabokrough et al, 2014). A research study published in 2015 suggested that an organometallic complex of platinum (II) azidothymidine (Pt (II)-AZT) has an antitumor effect on rat hepatocellular carcinoma cells. It was shown that this complex was significantly more effective in tumor suppression than the AZT without platinum (Sabokrough et al., 2014). Several organometallic complexes of AZT (with Zinc, Cobalt, Copper and Iron) were synthesized and characterized previously. The Iron complex of AZT (Fe-AZT) was shown to be the most stable (Shirvastav et al.) and have antimicrobial activity against 4 groups of bacteria. Electron rich ligands like AZT effectively bind and interact with metal ions producing metallodrugs which offer promising therapeutic application in terms of combating the drug-resistant strains of pathogens. It is also logical that a metal ion can influence the biological activity and therapeutic efficacy of the bio-molecule they bind (Shirvastav et al.) Thus it is useful to investigate the effect of a metal ion on the efficacy and mode of action of AZT in suppressing malignant cells and inducing apoptosis. To date, no research has been published regarding synthesis of Pd-AZT the inhibitory effect of both Fe-AZT and Pd-AZT on malignant cells. The aim of this research project is to synthesize Fe-AZT and Pd-AZT, confirm their structure and molecular mass and test their effects on viability of human hepatocellular carcinoma cells and normal human hepatocytes. Background Normal cell cycle In order to understand the biology of cancer it is crucial to understand the cell cycle of normal cells and cancerous cells. All cells in a given organism are under strict control of multiple regulatory agents, such as RB or p53 that control cell growth and keep the proliferative index stable. If there is a need for the cell to divide, the proliferative genes will turn on, or the suppressor genes that keep proliferation from occurring will be shut down, which will result in cell division (Hardin et al., 2014). The normal cell cycle consists of the following phases: G1, S, G2, M. During the G1 phase duplication of organelles, membrane systems and other important components happens. There is an important checkpoint at the end of this phase referred to as the restriction checkpoint, which verifies that DNA synthesis was successful and no errors have been detected. Next is the S phase during which the cell duplicates each chromosome. During the G2 the rest of the components that were not synthesized in the G1 phase are synthesized. A checkpoint after this phase makes sure the cell is ready to divide, and no errors in chromosome duplication were made. Mitosis is the next phase, when the cell physically divides (cytokinesis) making two identical daughter cells. A checkpoint during this phase checks if both of the daughter cells received the correct number of chromosomes. After division, the cell may remain in its silent G0 phase, when it does not divide, but continues to function. If any errors are detected on any of the checkpoints, the cell cycle is put on hold and the cell will undergo apoptosis. This process is governed my many enzymes and complexes, and theoretically, malfunction of any of these enzymes may result in uncontrolled cell division/proliferation. Cancer cell cycle It is important to note that the normal cell cycle is under control of thousands of regulatory elements that are coded for by different genes. If mutations occur in the genes that code for the key elements keeping cell proliferation at bay, one level of control over the cell cycle is lost. There are cell signaling mechanisms through which immune cells detect faulty cells, send them death signals and the faulty cell undergoes apoptosis. It takes more than one mutation for a normal cell to start behaving like a cancer cell. Usually it is either the loss of the function of suppressor genes, or overexpression of proliferative genes that result in uncontrolled cell proliferation. When the cell becomes malignant, it loses the ability to respond to death signals, thus does not undergo apoptosis. The malignant cell cycle is slower than the normal cell cycle, however, since the cancer cells keep dividing and do not die, their number grows exponentially resulting in tumors (Weinberg, 2014). There is distinct cytological difference between normal and cancer cells (Figure 2). A normal cell has a smaller, regularly shaped nucleus, a low nucleus/cytoplasm ratio, is well differentiated and has well defined borders. Cancer cells have larger, irregularly shaped nucleus, high nucleus/cy toplasm ratio, are less differentiated and have irregular borders. In contract with normal cells that adhere to each other, cancer cells are less adhesive and break away from each other (Weinberg, 2014). Figure 2. Azidothymidine (AZT), Fe(III)AZT, Pd(II)AZT Azidothymidine (AZT, Zidovudine) is a thymidine derivative in which the 3-hydroxy group is replaced by an azido group (Figure 3). It has been shown to have antitumor effects on different animal carcinoma cells both in vitro and in vivo through inhibition of telomerase activity and by causing cell cycle arrest (Gomez et al., 2012; Hadizadeh et al., 2014, Cooper and Lovett, 2011; Matteucci et al., 2015). Since cancer cells have higher proliferation rate than normal cells, their thymidine turnover rate is higher, which could contribute to their increased sensitivity to AZT. Several studies conducted by Fang et al., have shown that a hepatocellular carcinoma cell line (HepG2) is significantly more sensitive to AZT toxicity as compared to the normal hepatocyte (THLE2) sensitivity (Fang et al., 2014; Fang and Beland, 2009). The current hypothesis on how AZT affects the cell suggests that AZT is phosphorylated intracellularly yielding AZT-triphosphate and AZT-monophosphate. The AZT-monophosphate can be incorporated into the DNA structure instead of thymidine due to its similar molecular structure and shape. However, in contrast with thymidine, AZT lacks the 3 hydroxyl group which is the group that forms phosphodiester bonds between nucleotides in the DNA backbone structure. This means, wherever the AZT phosphate is incorporated in one of the DNA strands during DNA replication, the elongat ion of that strand halts (Figure 4). This is one of the mechanisms through which AZT causes damage to DNA (Fang et al, 2014). The commercial name of AZT is Zidovudine, and it is the basis of AIDS treatments. AZT blocks the replication of HIV-1 virus by competitively inhibiting the viral reverse transcriptase (RT). In other words, HIV RT prefers AZT to normal nucleotides. As described above, AZT is phosphorylated to AZT-triphosphate and is incorporated into viral DNA, halting nucleotide chain elongation. It is also known that AZT-triphosphate can be incorporated into eukaryotic DNA, although its affinity for DNA polymerases is lower than that for RT (Gomez et al, 2012). It was shown that Pt(II)-AZT is more effective in suppressing cancer cells than AZT, thus it has been suggested that having a transition metal bound to the central nitrogen atom of the azido group increases the complexs affinity for G-C and A-T base pairs and the P-backbone of DNA (Das and Pitre, 2007; Sabokrouh et al, 2015). Materials and Methods Fe-AZT and Pd-AZT synthesis To carry out the synthesis of Pd and Fe complexes, solid AZT and a standard ion solution of each metal (iron nitrate nonahydrate (Fe(NO3)3 ·9H2O and palladium chloride (PdCl2)) will be purchased from Sigma Aldrich (St. Louis, MO). Synthesis will be performed according to the protocol by Das and Pitre (2007). A 1:1 molar ratio of aqueous solution of AZT and metal ion will be refluxed for 3 hours. The volume of the reaction mixture will be reduced by 75% in order for the complex to precipitate. The solid product will be vacuum filtered, washed twice with ice-cold water, recrystallized and air-dried overnight. Infrared absorbance spectra will recorded for the AZT, Fe-AZT and Pd-AZT complexes. In the experiment conducted by Das and Pitre (2007) the comparison of IR spectra of pure AZT and its complex with Fe, the spectrum indicated a shift in bands from 2170 to 2150 cm-1 due to complexation through N atom of azido group. A similar band shift in the IR spectrum of Fe-AZT and Pd-AZT is expected. A Matrix Assisted Laser Ioniation/Distortion Time-of-Flight (MALDI TOF) mass spectrometry analysis will be performed for the Fe-AZT and Pd-AZT to determine the molar mass and the stoichiometry of the complexes. The matrix for analysis will be composed of 1:1 saturated Anthranilic Acid and Nicotinic Acid (Sigma Aldrich, St. Louis, MO). The samples and matrix will be dissolved in 45% acetonitrile: 55% water (van Kampen et al., 2004). Cell cultures HepG2, THLE2. Liver carcinoma cell line HepG2 and normal human liver cell line THLE2 will be obtained from American Type Culture Collection (Manassas, VA). The standard protocol recommended by ATCC will be used for establishing and maintaining hepatic cell lines. The cell lines will be plated at a density of 5 x 103 cells/cm2. HepG2 cells will be cultured in DMEM with 10% fetal bovine serum and antibiotics (penn/strep). THLE2 cells will be cultured in LHC-8 medium with 70ng/ml phosphoethanolamine, 5ng/ml epidermal growth factor, 10% fetal bovine serum and antibiotics (Fang et al., 2009). After ensuring sufficient cell growth and several passages of each cell line, the two cell lines will be divided into 4 groups each: control (no treatment) and treated with each drug: AZT, Fe-AZT and Pd-AZT (Table 1). HepG2 cells will be incubated with 2, 20 or 100  µM aqueous solution of AZT, Fe-AZT or Pd-AZT for 14 days. THLE2 cells will be incubated with 50, 500 or 2500  µM aqueous solution of AZT, Fe-AZT or Pd-AZT for 14 days. Each group of cells will be seeded at 5 x 103 cells/cm2 density in 6-well plates. The cells will be passaged weekly during the two-week treatment period. The dosage and incubation time were chosen based on a similar study (Fang et al., 2013; Matteucci et al., 2015; Sabokrouh et al., 2015). Cell viability assay After 14-day treatment period, 103 106 cells from each group will be seeded in a 96-well plate, incubated for 4 hours, and treated with MTT reagent, followed by a 8-12 hours incubation at 37 °C. After the incubation with MTT, when the cells have metabolized the yellow tetrazolium dye (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) to purple formazan, the cells will be treated by a detergent to release the formazan into the solution, and the absorbance will be measured at 570nm using an ELIZA reader. The number of metabolically active cells will be determined using a previously made calibration curve. The statistical differences will be calculated using a Two-Way ANOVA and Tukeys Post-Hoc Test. Anticipated outcomes. The expected mass-to-charge ratio from mass spectrometry analysis of Fe-AZT complex is 323.1, that of Pd-AZT is 373.6. Based on the published literature and the adopted hypothesis, all three drugs are expected to decrease cell viability. The concentration of either drug should have positive correlation with decrease in cell viability. Based on previous observations, it is expected that the HepG2 cells will be significantly more sensitive to both complexes, thus will be significantly less viable after the treatment compared to the THLE-2 cells. Study Participants and Timeline Anna Harutyunyan is the primary author of this study. She is currently a senior biology and chemistry major at Wilson College. This project will serve as Annas senior research project. She will conduct the research and present the results with the supervision of her advisors Dr. Deborah S. Austin (Sponsoring PAS member) and Dr. M. Dana Harriger (PAS Member). The research study began in September 2016 and will be completed by April 2017. Annas research findings will be analyzed and written as her senior thesis. Anna will also present her findings at the 2017 Pennsylvania Academy of Science meeting and at the Wilson College research colloquium. The Triangle of Death, The Limbo of the Lost, The Twilight Zone, The Port of Missing Ships, The Devil’s Triangle, The Hoodoo Sea, The Deadly Triangle. These and many more are names for the mysterious Bermuda Triangle. The Bermuda Triangle has been a huge mystery to many since the early 1900's. The Triangle is an area located in the western Atlantic Ocean, where supposedly many ships, people, planes, and other objects disappear unexplainably. The three points of location that connect the triangle are Miami, Florida, Bermuda, and San Juan, Puerto Rico. The triangle covers about five hundred thousand square miles of ocean. However, there is no official boundaries or markers for the triangle, just recognized linings. Over the past five centuries, more than a thousand ships and planes have disappeared inside of the triangle boundaries, and still to this day continue to do so. There are many theories behind these happenings. However, no one has yet found the real truth behind it. One of the most famous, or well-known happenings of the Bermuda Triangle is the disappearance of Flight 19. This happened in December, 1945 during a US Navy training exercise. Fourteen crew members on a route mission disappeared after sending out many radio messages, and when two Martin Mariner rescue planes came to search for them, only one came back while the other strangely could not be found. It is still not fully known what went wrong here. Many wrecked parts from Flight 19 have never been recovered. More famous incidents of the Bermuda Triangle are: The USS Cyclops and three hundred and nine crew members went missing in 1918, the Douglas DC-3 plane with thirty-two people that went missing in 1958 (no trace of aircraft was ever found), and a yacht that was... ...iangle disappearance is no more logical than trying to find a common cause for every automobile accident in Arizona.† Neither maritime insurance leader Lloyd’s of London, or the U.S. Coast Guard, do not see the Bermuda Triangle as an especially dangerous place. They say, â€Å"In a review of many aircraft and vessel losses in the area over the years, there has been nothing discovered that would indicate that casualties were the result of anything other than physical causes. The ideals of Hitler, a man wanting to eliminate all Jews and minorities, are views that can be questioned. Most people have difficulties accepting failure, and when failure arises people look elsewhere to blame someone else for their shortcomings. In society these people are called scapegoats. The question that comes to mind is Who was Hitler and what were his thoughts?   Ã‚  Ã‚  Ã‚  Ã‚  Most prominent leaders that can be thought of as great dictators were extremely forceful. Julius Caesar and Napoleon both rose up during troubling times of their country. They did this by going to war and winning, they would take over a country, set up a government, and then move on. Hitler was the exact opposite, he was a barbarian who ruled by striking fear into people. â€Å"They regard me as an uneducated barbarian. Yes, we are barbarians! We want to be barbarians! It is an honorable title.† (Rauschning, Hermann. The Voice of Destruction New York, 1940, p.80) The government that Hitler proposed had no real law. The people were to follow what they were told by those who were ranked above them, thus always leading back to Hitler. Hitler taught children to spy on their parents, and then report their wrong doings. He wanted people to hate their neighbors. He wanted those people of different, and for that matter â€Å"wrong† religions, political party, and/or race to be eliminated. A lot of the time when people look at history books and see a leader such as Hitler they might think that it was a long time ago, or that people were not that smart. He preached that before a government can reach a victory or a happy state it must first undergo a change. This is why the nazi movement was necessary. Make and Recieve Telephone Calls

The telephone that I use for work is a complicated phone and I don’t use all of its features, the basic features it has are the function to answer the phone and transfer calls to another person, it also allows me to answer a call from some else’s phone that is in my group. To transfer calls to another colleague I have to press R on the phone and then enter my colleague extension number for the call to be transferred. If I wanted to answer another colleagues phone I simple have to press *84 on the keypad and pick up the phone to receive their call.I am able to adjust the volume of the ring and I am able to place the call on loud speaker. I am also able to turn on or off the microphone in the handset as well as view the list of the numbers that I called last. I am also able to create a directory of number on the phone to be able to make calls quickly, on top of this there are 12 preset phone numbers with quick dial buttons on the phone however the previous owner had set t hese up and I am unable to change them.The phone is also capable of diverting the call to another phone and send messages between phones although I haven’t used this yet. Before making a phone call it’s a good idea to identify the purpose of the call as it would make me the caller sound more professional as I know exactly what I want and would waste no time with anything else, it would also allow me to have information that I can then use to prepare questions to ask and get the necessary details which I may require.An example of this is when a colleague requested that I acquire some information regarding the prospectus of the local colleges I had to identify whom I was going to make the call to and what the call would be regarding, after speaking to my colleague more and gaining the knowledge of exactly what they wanted me to get I was able to identify that the purpose of this call was to contact the college and ask what lessons they would be providing that relate to t he request made by my colleague.There are multiple ways to acquire the telephone number of people in my organisation as I can use the local address book to get there details, for example if I only knew there phone number which within the organisation would only be their extension number, I could use that to acquire their name and location as well as the department they work in, and I could do the same with only their name. Another method of gathering the contact details of someone is either using the internet to look up their business and get the information from their or ask a colleague if these have access or know the individuals name or number.I was request to book a venue for a stakeholder workshop for a colleague and they provided me with the specifics of the workshop but not where the workshop would be, they left that up to me to decide, therefore I had to determine which venue would best suit the requirements made. I spoke to another colleague that recommended Dillington Hous e, however they were unable to provide any contact details for me. Due to Dillington House not being within the organisation I was unable to use the local address book so I had to use the internet to acquire their contact numbers.After locating their website via a Google search I found the contact details for a Robin Kyd who arranged bookings. To finally make the call to the individual within the organisation I would simple enter their extension number on my phone and the call would be put through. On the other hand if I need the contact details of someone outside the organisation I would either have to research their details on the internet, this would include looking up their business, and getting their number from their website.However this number may only be a general enquiry number and I would then contact them and ask for the relevant details I need. However to make the phone call to someone outside the organisation I simply have to enter the number 9 before entering their ful l phone number to make the call. An example of me doing this is when I had to make a phone call to Dennis Quick who works for Taunton Deane, I was given his contact details from a colleague when they passed a message along to me.When I first made the call the line was engaged and I couldn’t get through, but when I rang again later that day I reached Dennis’s secretary who said he was available and transferred my call along to him, it was confirmed that I was speaking to the correct person when he introduced himself as Dennis Quick. The purpose of giving a positive image of myself and my organisation is to give me a polite, efficient and professional image otherwise people will not respect or take you seriously and if you have a professional image then this reflects well on the organisation.If you have a positive image then communication between people both within and outside the organisation would be better and more efficient possibly allowing for the transfer of infor mation quicker and efficiently. Summarising a telephone conversation before the call ends can be beneficial to both parties as it allows everyone to agree on what has been said and ensure there are no confusions this is especially useful when the telephone conversation has been going on for a long period of time and various things have been discussed, its easy to lose focus on what has previously been said and summarising overcomes this.It also allows objectives to be clear and information fresh in the mind for both parties to write down or relay to another person or persons. An example of this is when I had to make alterations to a booking that I had made with a business called Monks Yard in Ilminster. After discussing with the person that was arranging the meeting on their end, I ended the telephone conversation by asking the person to repeat the amendments that I wanted them to make and once I was happy that he had everything correct by checking with the list that I had in front of me at the time, I thanked them and ended the call.When a call comes into the office I’ll always take a name (full if possible and preferably) and the company they are calling from, I’ll try and obtain from the caller either what the problem is exactly or from which department the problem arisen from and if they know who within the department they spoke to. I’ll pass messages along to departments or relevant staff members and see what information can be found to resolve the problem.If this is not possible try and locate a member of staff who can help from the information obtained from the caller e. g. A company was expecting a Goods Receipt Note via SAP a week ago, I logged into SAP and saw that I needed to send the Goods Receipt Note but as I didn’t have an training and didn’t know what to do, I located a staff member who I knew had training on this and could help me, I contacted her department and was put through to her and she talked me throug h the problem, however I hadn’t been set up correctly and couldn’tsend out a Good Receipt Note and had to contact IT and get it fixed, during this entire period of getting it fixed and finally sending out the GRN I kept the company informed on what happened and gave them an estimated time that they would receive the GRN. The organisational structure of County Hall is that of a hierarchical structure which uses clear reporting lines and it is easy to see what each team is called and how many people are in that team and how they relate to other people in the system.The communication channels for us are different per team or department for me my phone is linked with everyone else’s in my team and I am able to answer their phone by entering *84 however I am not able to answer any other phone on the floor this way. I can contact anyone internally as long as I know their extension number as this is the main method of contacting anyone internally and I can call anyone externally by simply entering the number 9 before entering their phone number.When I make a external call I introduce myself by saying my name and company that I’m calling from, if I know prior to the phone call which member of staff or department that I want to call I would request to speak to them. If I don’t know who I’m reaching I would explain why I’m calling and request help on locating the member of staff that can help me. When I’m connected to this person, I ideally should have some questions ready or know exactly what information I require.At the end of the call I should summarise the call and recap information to make sure that it is accurate. If a fault was to occur with my telephone I would simply call the IT department and inform them when the problem occurred and how frequent it is happening, they would resolve the issue by either providing a replacement handset until it is fixed or locating the fault and fixing it at their earliest c onvenience.When I received a call I should identify who the caller is and their needs to do this I should follow my phone call procedures and gather the callers name and who they calling from, with this done I have now identified my caller and go about identifying their needs. Sometimes the caller will not provide you with their name at the start of the call as for example in a telephone conversation I had with a man asking for a colleague who was away from the desk, after following call procedures I asked to take a message, once the caller had given me his message, I asked who the call was from and he provided me with his name.Following procedures I will then ask the caller what I can help them with and they should briefly explain why they are calling, depending on why they have called and what my job entails I will either transfer them to another member of my team who can deal with their call, check the availability of a team member and inform the caller that they will call them b ack at a certain time or providing the information that they require to the best of my ability, although I will inform them that I am new and may not be able to answer all their questions.An example of this is when I received a phone call from a lady who wanted to submit a complaint to our department, after taking her name and contact details, she proceeded to tell me why she was complaining and what she would like done about it.During this telephone call I was taking notes so I could pass along her message when the call had ended, at the end of the call when she had finished explaining the problem, I repeated to her the points that I had taken and asked if these were correct, after making some small arrangements I summarised the information that she had given me and once she confirmed it was all correct, I informed her that I was going to pass this along to the relevant person and he would respond as soon as he can, I also informed her that I would call her when I had done this so she would know that action had been taken and it would also create a positive image of myself and my team. Giving accurate information to callers means when someone calls they have done so with the anticipation of getting their question or query answered promptly and professionally. Having up to date information available in a clear and concise manner will confirm in the callers mind that they question has been answered properly. For example if someone rang up asking for details on the most recent consultation regarding one of the teams projects and I gave them a out of date or incorrect document details then it would make a bad impression for the team and could affect their reputation as they may be deemed unreliable.From a callers point of view they want their personal details kept secure and confidential by showing this when dealing with a caller I am showing them that I can be trusted with their personal information and from my point of view, I need to keep the information priva te and respect their privacy otherwise I could become liable for prosecution under the Data Protection Act and with the information that my team handles a lot of it is private and confidential so this is a daily thing. If any confidential or information to do with security is given out, then it is causing a threat to others. Types of information that could affect confidentiality and security are people’s personal details so in my case it would be personal phone numbers and addresses. Therefore to reduce the risk of breaking confidentiality the information should only be handled by those that absolutely need access to it and if anyone requests the information then I should ask the persons whose information it is if they can have it or inform the person asking that someone will contact them.The Data Protection Act was setup to prevent confidential and security information from being given out and it is breaking the law if I was to do so. All these sorts of information should be handled by a maximum of 2 people and seen by no one else; this way no information will be revealed. After answering the call following the team’s procedure, I’d ask the caller if they know the name of the person they wish to speak to, if they don’t ask regarding which department. If they caller doesn’t know who to speak to then I would find out briefly, as they call may be regarding a private matter, what the call in about. Using the information I had gathered it would make it easier to transfer the call to the right person or department.If the person the caller is trying to reach is unavailable either because they are out of the office or away for the moment, then I would try to located someone else that may be able to take the call and explain to them the name of the caller, and what they are calling about as this would save time for the caller as they wouldn’t have to relay the information again. A lot of the time the case with transferring a c all is that the caller would either leave a message or inform me that they would call later, however with some calls that I receive the person they are trying to reach have left the desk momentarily and return while I’m still talking to the caller allowing me to transfer the caller to the person they were trying to reach. I would then inform the caller that I am transferring them to that particular person and explain why they would be more useful and helpful to deal with their request.Another option would be to leave a message I would either leave a message on their answering machine or a note on their computer, I would leave the following details; who the caller was, full name, where they were calling from, the time that they rang and the details of the call and any specific details or requests the caller wanted the intended receiver to know. A problem that could occur and has occurred it when transferring a call to a colleague the connection to the caller has become lost du ring the transfer, To amend this I would simply call them back apologising for the inconvenience, also ringing them back would show that I am keen to help them and am trying my best to provide good customer service. I would then log the problem as a technical fault, however if the problem kept occurring then I would report the problem with the IT department.Another problem that could occur is that I may transfer the caller to the wrong person this may have happened due to a system fault or a technical fault, though in my case it could be due to the fact that I’m new and could have made a mistake. If the caller went to the wrong number then the call would circulate around back to me and I would deal with the call accordingly. However if a call had been transferred to me by mistake then I would explain to the caller that unfortunately they had been transferred to the wrong person, though not necessarily the wrong department. I would apologise to the caller and ask for them to e xplain the problem and then transfer them to the correct person.I would then take a note of the problem and if the problem continued I would contact the transferee informing them that they have transferred the call to the wrong person and if it persists then I would inform IT about the problem. When receiving a phone call from an individual I would follow my teams phone call procedures this would involve doing the following things. If it was my phone that was ringing I was answer it as promptly as possible with the greeting, â€Å"Isaac Young, Major Programmes†, If it was another member of my teams phone that was ringing and due to the layout of the office it is hard to tell which phone is ringing, It is procedure to wait for the phone to ring three times and if it hasn’t been answered by then, I would enter *84 on the phone to take the call and answer with the previous greeting.Once I am connected to the caller I would obtain who the caller is and where they are from, by now the caller has usually mentioned the person they wish to speak to, if the caller mentions why they are calling this is noted when transferring the call and it is not company procedure although it is recommended that you ask what the call is in reference to. When reporting a fault to the IT department I have to be as exact as possible as this will help the issue to be resolved as fast as possible. Having an accurate account of when the problem first started and the frequency the fault occurred, what is actually happening, which can be obtained by the fault log that I have written down. I would request IT to provide a replacement phone while the problem is amended.
